Two sites on WDR5 mediate most of its interactions

Major interaction sites on WDR5. WDR5 interacts directly or indirectly with almost 200 proteins. Yet direct binders contact WDR5 through just one of two sites; either the “WBM” or the “WIN” site. The reuse of these same sites on WDR5 is likely important in enabling WDR5 to manage its many functions in the cell, but raises the question of why WDR5 has its fingers in so many biological pies. Perhaps discovery of potent WBM and WIN site inhibitors will help answer this question?
